The Allergenicity of Airborne Pollen at Pasar Minggu, South Jakarta in Wistar Rat (Rattus norvegicus)

(Indonesia), Master of Science in Biotechnology (Bogor Agricultural University)

Thesis Abstract:

Allergy cases were frequently reported in Kemacatan Pasar Minggu, South Jakarta. Pollen is one of the respiratory system allergens, which has never been explored in Indonesia. This research was conducted to know the allergenicity of airborne pollen in Pasar Minggu, South Jakarta. The protein of airborne pollen in the area was identified and characterized. The detection of allergenicity was made to rat (Wistar strain) by in vivo experiment. There were four kinds of airborne pollen found in Pasar Minggu, South Jakarta--maize, dwarf coconut, pine, and oil palm pollen. SDS-PAGE results showed that maize, dwarf coconut, pine, and oil palm pollen had several proteins with molecular weight ranging from 17-70 kDa, 17-68 kDa, 16-68 kDa, and 15-68 kDa, respectively. Skin inflammation was detected after injection of the protein extract to mouse’s subcutaneous. Respiratory challenge caused an increase in relative weight of respiratory tract. This challenge, though caused a decrease in body weight and an increase in size of mucous membrane cells, a secretion of mucous, and an eruption of inflammation cells. All pollen were allergenic to Wistar rat, thus can be utilized as an extract allergen for allergy detection and immunotherapy substances for humans in Indonesia.
